正宗台灣十六張麻將一共提供了自由對戰、修業模式、麻將大賽等三種模式，也有一些特殊的道具和法術可以使用，增加了不少趣味。喜愛麻將的玩家一定會很喜歡這款遊戲。

某天，師父給了主角兩萬元現金及兩張招待卷，要主角出外歷練麻將技能，以
參加賭國至尊盃麻將大賽，一雪師父前次敗北之恥，並重振風雲麻將館的聲威。
自由對戰：
從十六位人物裡選出自己與其他三位對手對戰。
修業模式：
從十四位人物裡選出代表自己的人物，進行劇情故事。
麻將大賽：
從十六位人物裡選出代表自己的人物進行麻將比賽。歷經初賽、資格賽、晉級賽、預賽、複賽、總決賽，前五場取前兩名晉級，最後一場以賭資最高者獲得優勝。
1.麻將館
(1)回房休息：恢復體力與精神力，若身上金錢不夠，且無物品時，師父會再補足為12000元。
(2)技能鑑定：經驗值足夠後可進行鑑定升級。必須答對兩題才能升級，若答錯則升該級的經驗值會再提高，故鑑定前要先存檔。
2.遊樂場：
每次消耗一張招待卷，隨機玩下列賭博遊戲：
(1)雙龍搶珠擲骰子拿走對應點數的麻將牌組合，直到無法拿為止，計算剩餘牌點數，少者獲勝。
